Encore largement méconnu, il est l'un des pères de l'informatique en ayant développé le premier calculateur électromécanique Z1 en 1938. La plus grande de ses réussites fut peut-être d'avoir réalisé le premier ordinateur du monde le Z3 en 1941. Celui-ci fut détruit en 1944 par les bombardements alliés et servait à produire des calculs pour une usine aéronautique allemande. La démarche de Zuse fut davantage celle d'un ingénieur isolé recherchant une solution pratique, que celle d'un théoricien. Ceci le conduisit à être effectivement le premier programmeur d'un calculateur automatique physique. Il le conçut sur la base du Système binaire inventé par Gottfried Leibniz. Il est probable qu'il connaissait les travaux de George Boole sur l'algèbre binaire (Booléene) et de Claude Shannon qui proposait d'appliquer cette algèbre aux relais téléphoniques afin d'obtenir des machines logiques. Zuse eut des résultats semblables aux travaux de Charles Babbage alors qu'il ne connut (voir discussion) ses travaux qu'après la fin de la guerre. Bien qu'il eut l'idée d'en intégrer dans son Z4, il ne put recevoir des nazis des tubes à vides trop précieux.
