A dominant theme of research in my laboratory during the past ten years has been the development and application of recombinant DNA and related methodologies for the analysis of genetic recombination in eukaryotic cells.
An important goal in cell and molecular genetics of mammalian cells is to be able to target exogenous genes to specific loci in the genome. This has been achieved although the frequency is low. Our current program aims to discover more about the process of gene conversion, a non-reciprocal recombination process in which sequences from one duplex DNA replace a corresponding region in a homologous duplex DNA. Both in vitro and in vivo experiments are being pursued to explore this reaction, particularly the isolation of yeast mutants blocked in their ability to carry out gene conversion between episomal and chromosomal homologous sequences. We are exploring this strategy for targeting alterations in chromosomal genes of embryonic stem cells with a goal of creating mutant animals.
The HIV-1 virus induces an immunodeficiency disease in patients it infects by depleting the T-helper population. We are exploring the signal transduction pathway from the T cell receptor as a possible target for the action of one of the viral genes.

Stanford Report, October 3, 2001 Paul Berg Paul Berg, the Robert W. and Vivian K. Cahill Professor of Cancer Research, Emeritus, and director emeritus of the Beckman Center for Molecular and Genetic Medicine; at Stanford 1959-present. Awarded the 1980 Nobel Prize in chemistry "for his fundamental studies of the biochemistry of nucleic acids, with particular regard to recombinant DNA." The other half of the award went to Walter Gilbert and Frederick Sanger "for their contributions concerning the determination of base sequences in nucleic acids." Since receiving the prize, Berg has continued to conduct research in the Department of Biochemistry, where his focus is the mechanism of repairing DNA damage. He continues to influence federal policy regarding stem cell research, biotechnology and human cloning.

Paul Berg Dr. Paul Berg was born in New York in 1926. In 1948, he received his B.S. from Pennsylvania State University and then received his Ph.D. from Western Reserve University in 1952. Dr. Berg is currently Willson Professor of Biochemistry at the Stanford University Medical Center. Dr. Berg's work in genetic research has earned him the title "father of recombinant DNA technology" and the 1980 Nobel Prize in chemistry. Berg was the first to splice together DNA from different species, an achievement which gave birth to a new fieldgenetic engineering. When Berg was awarded the 1980 Nobel Prize, he was cited for "his fundamental studies of the biochemistry of nucleic acids with particular regard to recombinant DNA." In one of his experiments he removed DNA from an animal virus called SV40 and inserted it into a circular DNA segment called a plasmid, which when introduced into a bacterium would have been reproduced. Concerns at the time regarding potential biohazards prompted Berg to stop short of carrying out the last step. Berg was one of the leaders in the scientific community in setting up structures for the safe, responsible continuation of such research. Since then scientists have transplanted DNA segments from one organism into another, usually bacteria, and thus created new life forms. The numerous honors Berg has received include being named California Scientist of the Year in 1963 and in 1981 co-recipient of the Albert Lasker Medcal Research Award.

During the thirty-five years that he resided in Kitchener-Waterloo he became the best-known musician in there region. As owner of a music centre, Director of music at Kitchener Collegiate Institute, University of Waterloo and Central Presbyterian Church in Cambridge Ont. Conductor of the Schneider Male Chorus, University of Waterloo Conference Co-ordinator, Theatre Administrator and Director of Creative Arts at the Cultural Program Centre, he gave of himself unstintingly in the furtherance of music and the theatre. He chairs the USB-IF Device Working Group, and co-chairs (with MCCI's Terry Moore ) the Communication Device Class committee. He has been involved with the development of USB technology since its inception. He recently was technical editor of the USB-IF Logo Compliance Testing test procedure document. Formerly Director of Engineering, USB Products at SystemSoft Corporation, he was directly involved with the development of PCMCIA technologies including CardBus, Zoomed Video, and Multi-Function cards. Paul has chaired several PCMCIA sub-committees; and served as the Technical Editor for the 1995 PC Card Standard. While working with PCMCIA he was a systems architect and software specialist at Ventura Micro, Inc. and at the Zenith Data Systems Advanced Technology Center in Santa Clara.

Stanley Rice Paul Berg witnessed firsthand the history of recombinant DNA research and regulation, having been in the forefront of both movements since he was a young man. He became a professor of biochemistry at Stanford University School of Medicine in 1959, when he was 33. He was elected to the National Academy of Sciences before he was 40, and he gained early recognition and influence when he delineated the key steps in which DNA produces proteins. Berg was awarded the Nobel Prize for Chemistry in 1980 for his work with DNA. In the mid-1970s, the National Academy of Sciences asked Berg to explore the safety of recombinant DNA technology. He responded with the historic "Berg letter," calling for a moratorium on recombinant DNA research until safety issues could be addressed. He was one of the key organizers of the international forum on recombinant DNA technology, the Asilomar Conference, which took place in February of 1975. One hundred leading scientists met at the conference to discuss the potential risks of gene-splicing experiments. The ensuing dialogue resulted in the National Institutes of Health guidelines published a year later, a milestone of responsible self-regulation in science.
