The Cutting Edge
Under Dr. Sly's leadership, a department actively engaged in modern biochemical research has emerged. Twenty talented faculty direct more than sixty students, fellows, and staff in research on fundamental problems in structural, cellular, and molecular biology. Specific areas of interest include protein and DNA structure and interactions, enzymology, regulation of translation, signal transduction, protein trafficking, and pathogen virulence. This research effort is supported by state-of-the-art research instrumentation and facilities. Keeping abreast of research and technological innovations is fostered by weekly departmental seminars featuring scientists from all over the world, a departmental journal club, and interactions with the extensive academic and commercial scientific community in St. Louis.
To the Future The department offers not only a diversity of research opportunities, but a faculty dedicated to providing outstanding training to scientists of the future. In an atmosphere conducive to learning, a spirited group of graduate students pursue programs tailored to their particular interests and needs. The department has always recognized that there is no conflict between academic training and the provision of marketable skills. Introduction to Enzymes
The use of enzymes in the diagnosis of disease is one of the important benefits derived from the intensive research in biochemistry since the 1940's. Enzymes have provided the basis for the field of clinical chemistry. It is, however, only within the recent past few decades that interest in diagnostic enzymology has multiplied. Many methods currently on record in the literature are not in wide use, and there are still large areas of medical research in which the diagnostic potential of enzyme reactions has not been explored at all. This section has been prepared by Worthington Biochemical Corporation as a practical introduction to enzymology. Because of its close involvement over the years in the theoretical as well as the practical aspects of enzymology, Worthington's knowledge covers a broad spectrum of the subject. Methods to Increase Bio-available Calcium Stores in Plants Expression of a high capacity Ca binding domain in plants for stress resistance and nutriceutical applications Calcium plays an essential role in both plants and animals, and modulation of cytosolic calcium levels is achieved in these organisms by a system of Ca -transport and storage pathways. Ca deficiency in plants can result in reduced growth or death of apical meristems, chlorotic or “burned” leaves, and increased susceptibility to plant pathogens. Losses caused by such effects are particularly important in many horticultural crops. At the same time plants provide a source of bioavailable Ca in our diets and there could be nutriceutical applications for plants with increased Ca levels Researchers at North Carolina State University (NCSU) led by Professor Niki Robertson have demonstrated the utility of using the high capacity Ca binding domain of calreticulin to increase Ca stores in plants. Arabidopsis plants expressing a fusion protein of the C-domain of maize calreticulin and the green fluorescent protein (GFP) showed:

Novel Bacterial Enzyme to Reroute the Phenylpropanoid
Pathway in Plants Pseudomonas HCHL gene and its uses in plants to produce novel flavour and industrial compounds Researchers at the Institute of Food Research (Norwich, UK) have identified and characterised a novel 4-Hydroxycinnamoyl-CoA Hydratase/lyase (HCHL) enzyme from Pseudomonas , which is capable of converting 4-coumaroyl-CoA, caffeoyl-CoA and feruloyl-CoA to the corresponding hydroxybenzaldehydes in vitro . These hydroxybenzaldehydes include vanillin, the major flavour component of vanilla. The substrates for this HCHL enzyme are intermediates of the plant phenylpropanoid pathway and therefore expression of the corresponding gene in plants has the potential to divert phenylpropanoid metabolism to produce novel metabolites. The approach could have wide ranging commercial utility since the plant phenylpropanoid pathway is responsible for the synthesis of a wide variety of important secondary metabolic compounds, including lignins, antioxidants, flavonoids and salicylates. As such these phenylpropanoids have an important effect on plant qualities such as texture, flavour, colour and processing characteristics. The phenylpropanoid pathway has also been the site of intervention for the production of bioplastics in plants. Initial data generated in transgenic tobacco plants and in hairy roots of Datura from the over-expression of this enzyme with the constitutive 35S promoter has demonstrated its ability to massively reroute carbon flux from the phenylpropanoid pathway into novel C

This competition is open to all third-level students (both North and South) who are interested in biochemistry. Submit a 1,500-word newspaper article, suitable for the non-scientist, explaining clearly and accurately the significance of any aspect of biochemistry/molecular biology in an engaging, informative and easily intelligible manner. The article may focus on international developments; however serious reference to relevant Irish research should be made. Reference to any commercial sponsorship for the work conducted in Ireland should also be included. Good quality graphics should be used to illustrate the article as appropriate. In order to facilitate publication, it may be necessary to edit the article.

    Much of the research in this emphasis group focuses on understanding how proteins are synthesized and assembled into functional macromolecules. State of the art biophysical technologies are exploited to define mechanisms for protein folding, and protein trafficking in the endoplasmic reticulum and nucleus. Teaching
The Department has teaching responsibilities on every year of the undergraduate MBBS Graduate Entry MBBS and BSc Biomedical Science courses, including lectures, tutorials, project supervision, organisation of modules and examinations. It also contributes to the course. The Intercollegiate MSc in Medical Immunology of London University is also co-ordinated from the Department. Professor Clemens is Course Director of the BSc Biomedical Science course and Professor Johnstone is Chair of Examiners. Dr Bashford is Vice-Dean of Undergraduate Medicine and Sub-Dean for Cycle 1 (Years 1 and 2 of the 5-year MBBS course). Professor Fisher teaches on the Intercalated BSc course in Tumour Biology offered by University College Hospital Medical School. Professor Hay is Chief Examiner and Chairman of the Board of Examiners for the MSc in Medical Immunology. Dr Poulton is Sub-Dean for Educational Technology with a major responsibility for web-delivery of teaching material. Dr Bashford is visiting examiner for Phase 1 of the undergraduate course at the Imperial College School of Medicine, Professor Johnstone is external examiner for the Royal Veterinary College (First BVet Med Part2) and Professor Clemens is external examiner for the University of Cambridge (2nd MB examinations). Several members of the Department also act as external PhD examiners at various universities. Professor Hay is External Examiner at University College London, for the MBBS, Basic Medical Sciences Parts I and II and teaches on the following external courses: Queen Mary and Westfield College - MSc in Clinical Microbiology; Brunel University MSc in Medical Genetics and Immunology; University of Surrey (Roehampton) MSc in Clinical Neuroscience and Immunology.
